Melanotan II is a synthetic cyclic 7-residue chemical sequence analog of alpha-MSH with non-selective activity at MC1R (tanning), MC3R (metabolic), MC4R (appetite/sexual function), and MC5R (exocrine). The cyclic lactam bridge between Asp and Lys residues enhances metabolic stability vs. linear α-MSH. Binding MC1R activates adenylyl cyclase → cAMP → PKA → CREB → MITF cascade, upregulating eumelanin synthesis in melanocytes.
