Semax is a synthetic ACTH(4-10)-Pro-Gly-Pro analogue that rapidly upregulates BDNF and NGF in the hippocampus and cortex. It activates MC4R (melanocortin 4 receptor) and TrkB (BDNF receptor), modulating dopamine D1/D2 signaling. The Pro-Gly-Pro C-terminal extension enhances metabolic stability against carboxypeptidases, extending intranasal effective duration to 30-60 minutes.
